美國製藥公司 Vertex Pharmaceuticals（Vertex）日前投入 12 億美元，攜手基因編輯巨頭張峰創立的 Arbor Biotechnologies（Arbor），透過 CRISPR 的技術開發體外編輯細胞療法，標靶糖尿病與血液性疾病。
開發最多 7 款藥物
本次合作中，雙方將開發最多 7 種藥物，適應症包含第 1 型糖尿病、鐮狀細胞性貧血（sickle cell disease, SCD）與貝他海洋性貧血（β thalassemia）。
Arbor 的創辦人之一張峰（Feng Zhang）博士曾與 2020 年諾貝爾化學獎得主 Jennifer Doudna 博士合作，是 CRISPR 基因編輯技術的研發團隊之一。在 2006 年創立 Arbor 後，他與研究團隊一共推出 CRISPR 在內的 7 種基因編輯工具。而這也是 Vertex 早於 2019 年 1 月就與 Arbor 合作的原因。
2019 年首次合作，標靶囊腫纖維化
2019 年首次合作，Vertex 與 Arbor 以尋找可基因編輯、且具高核酸複製忠實性（fidelity）與催化活性（catalytic activity）的 DNA 核酸內切酶（DNA endonucleases）為目標，來開發囊腫纖維化（cystic fibrosis）與其他 4 種疾病的基因療法。
Arbor 自身的產品線以肝臟疾病為主，其中原發型高草酸鹽尿症（primary hyperoxaluria, PH）療法已突破研究階段，正進行先導化合物優化。
Vertex 基因療法佈局
Vertex 在過去數年積極推動與基因療法公司合作，除了 Arbor 外，還有與 Jennifer Doudna 創立的 CRISPR Therapeutics、Affinia Therapeutics（Affinia）等公司。Vertex 更在 2019 年收購美國基因療法新創 Exonics Therapeutics（Exonics）。
回顧 Vertex 一連串在基因治療領域上佈局， 在今年（2021年）4 月擴大從 2015 年開始與 CRISPR Therapeutics 的合作案，投入 11 億美元深化基因療法 CTX001 的臨床與商業化進程。該療法可標靶 SCD 與輸血依賴型貝他海洋性貧血症（transfusion-dependent beta-thalassemia, TDT），目前正於第 1/2 期臨床試驗，也獲得美國 FDA 孤兒藥、再生醫學進階療法、罕見兒科疾病用藥等認定。
而在 2020 年 4 月，Vertex 啟動與 Affinia 基因療法公司合作，主要開發腺相關病毒（AAV）載體，來標靶裘馨氏肌肉失養症（Duchenne muscular dystrophy, DMD）、強直型肌肉萎縮症第一型（Myotonic dystrophy type 1, DM1）與囊腫纖維化。此合作案金額高達 16 億美元，其中 8,000 萬美元會在藥物研究階段就提供，以支援開發所需經費。
之前於 2019 年 6 月，Vertex 以 2.45 億美元收購 Exonics，加速 DMD 與嚴重神經肌肉疾病的基因療法研發，也支援與 CRISPR Therapeutics 的合作。
