美国制药公司 Vertex Pharmaceuticals（Vertex）日前投入 12 亿美元，携手基因编辑巨头张峰创立的 Arbor Biotechnologies（Arbor），透过 CRISPR 的技术开发体外编辑细胞疗法，标靶糖尿病与血液性疾病。
开发最多 7 款药物
本次合作中，双方将开发最多 7 种药物，适应症包含第 1 型糖尿病、镰状细胞性贫血（sickle cell disease, SCD）与贝他海洋性贫血（β thalassemia）。
Arbor 的创办人之一张峰（Feng Zhang）博士曾与 2020 年诺贝尔化学奖得主 Jennifer Doudna 博士合作，是 CRISPR 基因编辑技术的研发团队之一。在 2006 年创立 Arbor 后，他与研究团队一共推出 CRISPR 在内的 7 种基因编辑工具。而这也是 Vertex 早于 2019 年 1 月就与 Arbor 合作的原因。
2019 年首次合作，标靶囊肿纤维化
2019 年首次合作，Vertex 与 Arbor 以寻找可基因编辑、且具高核酸复制忠实性（fidelity）与催化活性（catalytic activity）的 DNA 核酸内切酶（DNA endonucleases）为目标，来开发囊肿纤维化（cystic fibrosis）与其他 4 种疾病的基因疗法。
Arbor 自身的产品线以肝脏疾病为主，其中原发型高草酸盐尿症（primary hyperoxaluria, PH）疗法已突破研究阶段，正进行先导化合物优化。
Vertex 基因疗法布局
Vertex 在过去数年积极推动与基因疗法公司合作，除了 Arbor 外，还有与 Jennifer Doudna 创立的 CRISPR Therapeutics、Affinia Therapeutics（Affinia）等公司。Vertex 更在 2019 年收购美国基因疗法新创 Exonics Therapeutics（Exonics）。
回顾 Vertex 一连串在基因治疗领域上布局， 在今年（2021年）4 月扩大从 2015 年开始与 CRISPR Therapeutics 的合作案，投入 11 亿美元深化基因疗法 CTX001 的临床与商业化进程。该疗法可标靶 SCD 与输血依赖型贝他海洋性贫血症（transfusion-dependent beta-thalassemia, TDT），目前正于第 1/2 期临床试验，也获得美国 FDA 孤儿药、再生医学进阶疗法、罕见儿科疾病用药等认定。
而在 2020 年 4 月，Vertex 启动与 Affinia 基因疗法公司合作，主要开发腺相关病毒（AAV）载体，来标靶裘馨氏肌肉失养症（Duchenne muscular dystrophy, DMD）、强直型肌肉萎缩症第一型（Myotonic dystrophy type 1, DM1）与囊肿纤维化。此合作案金额高达 16 亿美元，其中 8,000 万美元会在药物研究阶段就提供，以支援开发所需经费。
之前于 2019 年 6 月，Vertex 以 2.45 亿美元收购 Exonics，加速 DMD 与严重神经肌肉疾病的基因疗法研发，也支援与 CRISPR Therapeutics 的合作。
